| FOLR1

Folate receptor alpha (FOLR1) is a protein encoded by the FOLR1 gene. The protein encoded by this gene is a member of the folate receptor (FOLR) family. Members of this family exhibit high affinity for folate and several reduced folate derivatives, mediating the internalization of 5-methyltetrahydrofolate into cells. This receptor is responsible for binding folate and its derivatives, a function that becomes critically important during fetal development. The gene consists of seven exons; exons 1 through 4 encode the 5' UTR, while exons 4 through 7 encode the open reading frame. Due to the presence of two promoters, multiple transcription start sites, and alternative splicing of exons, the gene produces several transcript variants. These variants differ in the length of their 5' and 3' UTRs, but they encode identical amino acid sequences. Given the high expression of FRα in certain tumors, it represents an attractive therapeutic target for developing novel anticancer drugs that limit toxic side effects on non-target tissues.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 200 100 50 25 12.5 6.25 3.12 0
OD 2.36 1.49 0.95 0.54 0.35 0.26 0.21 0.09
Corrected OD 2.27 1.69 1.14 0.7 0.37 0.2 0.08 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human FOLR1 spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 84-101 96
Plasma(n=8) 92-105 102
Cell Culture Supernates(n=8) 96-108 104

| Linearity

Samples were spiked with high concentrations of Human FOLR1 and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4) Cell Culture Supernates(n=4)
1 : 2 Range (%) 84-95 88-96 90-110
Average (%) 91 93 96
